Preparation and Evaluation of Fast Dissolving Tablets Amisulpride Using Novel Super Distintegrants for The Treatment of Schizophrenia

The present research work was aimed at the preparation and evaluation of fast dissolving tablets of Amisulpride using novel super disintegrants to improve patient compliance and achieve a rapid onset of action in the treatment of schizophrenia. Amisulpride, an atypical antipsychotic, suffers from delayed onset of action when administered in conventional dosage forms; hence, fast dissolving tablets were developed by the direct compression method. Various formulations were prepared using different novel super disintegrants and were subjected to evaluation.
Pre-compression parameters such as angle of repose, bulk density, tapped density, compressibility index, and Hausner’s ratio confirmed good flow properties of the blends. Post-compression studies including hardness, friability, weight variation, thickness, and drug content uniformity were within pharmacopeial limits. The prepared tablets were further assessed for disintegration time, wetting time, water absorption ratio, and in vitro drug release. Results revealed that formulations containing novel super disintegrants showed faster disintegration and higher dissolution rates compared to those with conventional disintegrants. The optimized formulation exhibited rapid disintegration within seconds and achieved maximum drug release, ensuring faster therapeutic availability.
In conclusion, fast dissolving tablets of Amisulpride prepared with novel super disintegrants represent a promising approach for enhancing onset of action, improving bioavailability, and ensuring better compliance in patients suffering from schizophrenia.
