Formulation and Evaluation of Sustained Release Matrix Tablets of Theophylline Using Natural Polymer

The present study focuses on the formulation and evaluation of sustained release matrix tablets of Theophylline using natural polymers such as Guar Gum, Fenugreek, and Karya Gum. The aim was to develop a cost-effective and biocompatible drug delivery system that could provide prolonged therapeutic effects, reduce dosing frequency, and enhance patient compliance. A total of nine formulations were prepared using varying concentrations of the selected natural polymers by the Direct compression method.
All pre-compression and post-compression parameters, including hardness, friability, weight variation, thickness, and drug content, were found to be within acceptable limits as per IP standards, indicating the suitability of the formulations. In-vitro drug release studies were carried out for 12 hours, and the release profiles showed a sustained release pattern.
Among the nine formulations, F6 was identified as the optimized formulation as it exhibited a drug release of 99.72% at the end of 12 hours, following a controlled release mechanism. The results suggest that natural polymers can be effectively utilized for the formulation of sustained release tablets of Theophylline.
